چکیده

Objective: Activation of cholinergic muscarinic receptors leads to activation of membrane bound phospholipase C, leading to generation of IP3 and diacylglycérol, The present study aimed at measuring muscarinic receptor-mediated phosphoinositide breakdown in three different regions of rat brain. Methods: Cortical, hippocampal and striatal slices labeled with [3H] myo-inositol were incubated with muscarinic agonist carbachol (Cch) and LiCI. Reaction was terminated by the addition of chloroform/methanol and labeled inositol phosphates were extracted from tissue. Individual [3H]inositol phosphates were separated using anion exchange chromatography. Results: Cch caused a rise in IP, formation, where the increase over basal in the cortex, hippocampus and striatum were 98%, 73% and 45%, respectively. Conclusion: Phosphoinositide turnover was stimulated by carbachol in all brain structures studied, with the most pronounced effects observed in the cortex and hippocampus. Key W ords: Muscarinic receptors, inositol phosphates, carbachol, rat brain

Pertussis toxin does not inhibit muscarinic-receptor-mediated phosphoinositide hydrolysis or calcium mobilization.

Pertussis toxin was used to examine the role of the inhibitory guanine nucleotide regulatory protein, Ni, in muscarinic-receptor-mediated stimulation of phosphoinositide turnover and calcium mobilization. In cultured chick heart cells, pertussis-toxin treatment inhibited muscarinic-receptor-mediated attenuation of isoprenaline-stimulated cyclic AMP accumulation. This finding is consistent with ...

An M2 muscarinic receptor subtype coupled to both adenylyl cyclase and phosphoinositide turnover.

To investigate whether a particular receptor subtype can be coupled to multiple effector systems, recombinant M2 muscarinic receptors were expressed in cells lacking endogenous receptor. The muscarinic agonist carbachol both inhibited adenylyl cyclase and stimulated phosphoinositide hydrolysis. The stimulation of phosphoinositide hydrolysis was significantly less efficient and more dependent on...

Differential coupling of muscarinic M1, M2, and M3 receptors to phosphoinositide hydrolysis in urinary bladder and longitudinal muscle of the ileum of the mouse.

We investigated the coupling of muscarinic receptor (M) subtypes to phosphoinositide hydrolysis in ileum and urinary bladder using muscarinic receptor knockout mice. In urinary bladder from wild-type mice, the muscarinic agonist oxotremorine-M, elicited a robust phosphoinositide response characterized by an EC50 value of 0.22 microM and a maximal response (Emax) of 32.8% conversion of [3H]inosi...

Evidence that muscarinic cholinergic receptors selectively interact with either the cyclic AMP or the inositol phosphate second-messenger response systems.

The relative capacities of muscarinic cholinergic receptor (MR) and bradykinin (BK)-receptor activation to increase phosphoinositide hydrolysis and to increase cytosolic Ca2+ were compared in NG108-15 neuroblastoma x glioma and 1321N1 human astrocytoma cells. In 1321N1 cells, the muscarinic cholinergic agonist carbachol and BK each stimulated a concentration-dependent accumulation of inositol p...

Protein kinase C is involved in desensitization of muscarinic receptors induced by phorbol esters but not by receptor agonists.

Preincubation with receptor agonists or phorbol esters desensitized muscarinic-receptor-mediated [3H]cyclic GMP responses in mouse neuroblastoma N1E-115 cells. However, desensitization mediated by phorbol esters was heterologous, whereas that effected by receptor agonist was specific towards the muscarinic receptors. In addition, there was no loss of cell surface muscarinic receptors, as measur...
